Laboratory glassware and serial dilutions
- 3 things to know
- Components 01, 02 and 03
Overview
Recall questions on tenfold serial dilutions and doubling dilutions, calculating concentrations and the volumes needed, mixing and using fresh pipettes, volumetric flasks and making up a standard solution, reading a meniscus, dilution factors, and viable counts in colony-forming units.
What you need to know
Answer each one before you reveal it.
How is a tenfold serial dilution prepared from a stock solution?
How would you make 10 cm³ of a solution ten times more dilute than the stock?
Questions from this subtopic
Each one with the model answer underneath.
- 1
What is a serial dilution?
Model answer
It is a stepwise series of dilutions in which each solution is made by diluting the previous one by the same factor.
- 2
How is a tenfold serial dilution prepared from a stock solution?
Model answer
One part of the solution is added to nine parts of distilled water and mixed, and the process is repeated using each new solution in turn.
- 3
How would you make 10 cm³ of a solution ten times more dilute than the stock?
Model answer
1 cm³ of the stock is added to 9 cm³ of distilled water and mixed thoroughly.
- 4
What is a doubling dilution?
Model answer
Each solution is made by mixing equal volumes of the previous solution and the diluent, so the concentration is halved at every step.
- 5
Calculate the concentration remaining after three tenfold dilution steps of a 1.0 mol dm⁻³ stock solution.
Model answer
The concentration is 1.0 × 10⁻³ mol dm⁻³.
- 6
What volumes of 1.00 mol dm⁻³ glucose solution and water are needed to make 20.0 cm³ of a 0.200 mol dm⁻³ solution?
Model answer
4.00 cm³ of the stock is mixed with 16.0 cm³ of water.
