Culturing microorganisms in the laboratory
- 3 things to know
- Component 1
Overview
Two main physical forms of culture medium, four things a nutrient medium must supply and growth factors.
What you need to know
Answer each one before you reveal it.
What is agar, and why is it used as the setting agent?
Name four things a nutrient medium must supply.
Questions from this subtopic
Each one with the model answer underneath.
- 1
Name the two main physical forms of culture medium.
Model answer
Nutrient broth, which is liquid, and nutrient agar, which is solid.
- 2
What is agar, and why is it used as the setting agent?
Model answer
It is a polysaccharide extracted from seaweed that dissolves in hot water and sets to a firm gel below about 40 °C, and hardly any bacterium can digest it, so the surface stays solid while the culture grows.
- 3
Name four things a nutrient medium must supply.
Model answer
A source of carbon and energy such as glucose, a source of nitrogen such as peptone, mineral ions, and water.
- 4
What are growth factors, and why are they sometimes added to a medium?
Model answer
They are vitamins and similar substances that some species cannot make for themselves, and they are supplied by adding yeast extract or blood to the medium.
- 5
What is a selective medium?
Model answer
One containing a substance that prevents most species from growing, so that only the organism being sought forms colonies.
- 6
What is meant by a pure culture?
Model answer
A culture containing only one species of microorganism, with no others present.
