Light and electron microscopy
- 3 things to know
- A2 6
Overview
Magnification and resolution with worked calculations, oil immersion and phase contrast, transmission and scanning electron microscopes, sample preparation, and setting up a light microscope.
What you need to know
Answer each one before you reveal it.
Define magnification and resolution.
What is meant by the image and the focus of a microscope?
Questions from this subtopic
Each one with the model answer underneath.
- 1
Define magnification and resolution.
Model answer
Magnification is how many times larger the image is than the real object. Resolution is the ability to distinguish two points that are close together as separate.
- 2
What is meant by the image and the focus of a microscope?
Model answer
The image is the enlarged picture of the specimen formed by the lenses. The specimen is in focus when the lenses are positioned to give the sharpest image.
- 3
A cell 5 µm long appears 12 mm long in a micrograph. What is the magnification?
Model answer
The magnification is ×2400. The working is 12 mm = 12,000 µm, then 12,000 ÷ 5 = 2400.
- 4
An image is 30 mm long at a magnification of ×1500. What is the real length of the object?
Model answer
The object is 20 µm long. The working is 30 mm = 30,000 µm, then 30,000 ÷ 1500 = 20 µm.
- 5
How does oil immersion improve a light microscope's image?
Model answer
A drop of oil is placed between the ×100 objective and the slide. The oil has about the same refractive index as glass, so less light is lost by refraction and resolution improves.
- 6
How does phase contrast microscopy work?
Model answer
Light passing through different parts of a cell is slowed by different amounts. Special lenses convert these differences into variations in brightness, so unstained living cells become visible.
