Gene inactivation and knockout organisms
- 3 things to know
- Unit A2 2
Overview
Three ways in which a gene can be made inoperative, knockout mouse and knockin mouse.
What you need to know
Answer each one before you reveal it.
What is a knockin mouse?
Explain why inactivating a gene sometimes produces no visible change at all.
Questions from this subtopic
Each one with the model answer underneath.
- 1
Explain how inactivating a gene helps biologists to discover what it does.
Model answer
Removing the gene's product from the organism reveals what fails to work, so the change in the phenotype identifies the process that the gene normally controls.
- 2
State three ways in which a gene can be made inoperative.
Model answer
A gene may be disabled where it lies, removed from the chromosome altogether, or replaced by a defective version of itself.
- 3
What is a knockout mouse?
Model answer
A knockout mouse is a mouse in which a chosen gene has been made inoperative so that the effects of losing that gene can be studied in a living mammal.
- 4
What is a knockin mouse?
Model answer
A knockin mouse is one in which a chosen gene has been replaced by a different or altered version, so the effect of that particular variant can be studied.
- 5
Give three reasons why the mouse is used as a model organism for studying genes.
Model answer
The mouse is a mammal with organ systems and a genome very similar to our own, it breeds rapidly and produces large litters, and it is small and inexpensive to keep in numbers.
- 6
Explain how a knockout mouse can be used to study a human genetic disorder.
Model answer
The mouse version of the gene involved in the disorder is inactivated, and the animal then develops comparable symptoms whose progression can be followed from the earliest stage in a way that is impossible in a patient.
