The stages of gene transfer
- 3 things to know
- Unit A2 2
Overview
Sticky ends, how reverse transcriptase is used to obtain a gene and vector in gene technology.
What you need to know
Answer each one before you reveal it.
How does a restriction endonuclease help in obtaining a donor gene?
Explain why sticky ends are so useful in gene transfer.
Questions from this subtopic
Each one with the model answer underneath.
- 1
List the main stages of transferring a gene from one organism into another.
Model answer
The donor DNA is obtained, it is inserted into a vector, the vector is used to transform the recipient cells, the transformed cells are identified using a marker, and they are then cultured to give large numbers.
- 2
How does a restriction endonuclease help in obtaining a donor gene?
Model answer
The enzyme cuts the chromosomal DNA at specific recognition sites on either side of the required gene, releasing it as a fragment that can be handled separately.
- 3
What are sticky ends?
Model answer
Sticky ends are the short single-stranded stretches of unpaired bases left at each side of a staggered cut made by a restriction endonuclease.
- 4
Explain why sticky ends are so useful in gene transfer.
Model answer
The exposed bases will pair with any complementary single-stranded stretch, so a fragment and a plasmid cut with the same enzyme have matching sticky ends and join together readily.
- 5
Describe how reverse transcriptase is used to obtain a gene.
Model answer
Messenger RNA for the required protein is collected from a cell that makes a great deal of it, and reverse transcriptase uses that mRNA as a template to build a complementary DNA copy of the gene.
- 6
Give one advantage of obtaining a gene using reverse transcriptase rather than by cutting it out of a chromosome.
Model answer
The mRNA has already had its introns removed, so the DNA copy made from it contains only the coding sequence and can be expressed correctly by a bacterium.
