Combined spectroscopic structure determination
- 2 things to know
- Components 01, 02 and 03
Overview
Recall questions on combining infrared, mass spectrometry and NMR, and on working a structure out from a set of spectra.
What you need to know
Answer each one before you reveal it.
Why can mass spectrometry not usually decide between two isomers on its own?
Questions from this subtopic
Each one with the model answer underneath.
- 1
What does each of mass spectrometry, infrared spectroscopy and NMR contribute when a structure is being worked out?
Model answer
Mass spectrometry gives the molecular formula and the fragments present, infrared identifies the functional groups, and NMR shows the carbon skeleton and how the hydrogen atoms are arranged around it.
- 2
Why is it worth starting from the high-resolution molecular ion rather than the infrared spectrum?
Model answer
The molecular formula fixes how many atoms of each element there are, so it limits which structures the other two spectra need to be tested against.
- 3
A compound gives a high-resolution M⁺ at m/z 74.0366. Using ¹H = 1.0078, ¹²C = 12.0000 and ¹⁶O = 15.9949, show that its formula is C₃H₆O₂ and not C₄H₁₀O.
Model answer
C₃H₆O₂ gives 36.0000 + 6.0468 + 31.9898 = 74.0366, whereas C₄H₁₀O gives 48.0000 + 10.0780 + 15.9949 = 74.0729.
- 4
Its proton NMR shows a triplet at δ 1.1, a quartet at δ 2.4 and a broad singlet near δ 11.5, in the ratio 3 : 2 : 1. Identify the compound and say how all three techniques agree.
Model answer
It is propanoic acid, CH₃CH₂COOH: the formula C₃H₆O₂ allows it, the infrared confirms the acid group, and the triplet and quartet show an ethyl group next to the carboxyl carbon.
- 5
Why would a proton spectrum of two singlets in the ratio 3 : 3 have ruled propanoic acid out for that formula?
Model answer
Two unsplit signals mean no CH₃ and CH₂ are adjacent, which points instead to methyl ethanoate, where the two methyl groups have no neighbouring protons.
- 6
Why is a broad absorption near 3000 cm⁻¹ on its own not enough to prove a carboxylic acid?
Model answer
Alcohols and phenols also absorb broadly in that region, so a strong C=O absorption between 1700 and 1725 cm⁻¹ must be present as well.
