The Cell Cycle and Mitosis: Root Tip Squash and Mitotic Index
- 3 things to know
- AS Unit 1
Overview
Preparing a stained root tip squash to see dividing cells, the rules for scientific drawing, and calculating the mitotic index and the time spent in each stage of the cell cycle.
What you need to know
Answer each one before you reveal it.
Why is the tip of an actively growing root used to study mitosis?
Why is a root tip warmed in hydrochloric acid before it is mounted?
Questions from this subtopic
Each one with the model answer underneath.
- 1
Why is the tip of an actively growing root used to study mitosis?
Model answer
The meristem just behind the root tip is the region where cells are dividing rapidly, so many cells will be caught in the stages of mitosis.
- 2
Why is a root tip warmed in hydrochloric acid before it is mounted?
Model answer
The acid breaks down the middle lamella holding the cells together so that they separate and can be spread out.
- 3
Why is a stain such as acetic orcein used?
Model answer
It colours the chromosomes darkly so that they stand out against the rest of the cell and the stage of division can be identified.
- 4
Why is the coverslip pressed down firmly on a root tip preparation?
Model answer
Squashing spreads the cells into a single layer so that light passes through and individual cells can be seen clearly.
- 5
Give three rules that must be followed when making a scientific drawing of the cells observed.
Model answer
Use clear continuous single pencil lines with no shading, keep the proportions the same as those seen down the microscope, and add label lines drawn with a ruler that do not cross one another.
- 6
What must be recorded alongside a scientific drawing besides the labels?
Model answer
A title identifying the specimen and the magnification at which it was drawn.

What is inside Levo
Six ways to work the same material.
Every question in Genetic Information Is Copied and Passed on to Daughter Cells, on a schedule.
Attempt a question from memory, write the correct version, then attempt it again. Levo records the jump between your first attempt and your last, and brings each question back on the day you are due to forget it.
